修学旅行、合宿、初めて泊まるホテル。なぜか1日目の夜だけ寝つけない。これは「第一夜効果」と呼ばれ、1960年代から知られている現象です。
2026年9月、科学誌『Journal of Neuroscience』に、米国ブラウン大学(大阪大学の研究者も参加)のチームが、この現象を脳の化学物質のレベルで調べた論文を発表しました。すんなり眠れたときは、脳の前の方で「興奮系の物質」がスッと下がっていた。寝つけない初回は、それが下がっていなかったのです。
01「アクセル」と「ブレーキ」の物質を測る
脳の中には、神経の活動を高めるアクセル役と、しずめるブレーキ役の物質があります。
- Glx(グルタミン酸+グルタミン):アクセル役(興奮性)
- GABA(ギャバ):ブレーキ役(抑制性)
眠りにつくには「ブレーキを強く踏む」のか、「アクセルをゆるめる」のか。研究チームはこれを確かめるため、第一夜効果をわざと利用しました。
02寝つけた回だけ「アクセル」がゆるんでいた
予想どおり、1回目は寝つくまでの時間が長くなりました。そして脳内の物質を見ると、
- 2回目(眠りやすい回):起きている状態から眠りへ移る間に、Glxが低下
- 1回目(眠りにくい回):この低下が見られなかった
さらに、2回を通して、Glxの下がり方が大きい回ほど、早く寝ついていました。統計的な分析でも、1回目と2回目の寝つきの差は、Glxの変化で説明できることが示されました。
一方、ブレーキ役のGABAは、どちらの回でもはっきり変化せず、寝つきとも関係がありませんでした。

03「片方の脳が見張っている」研究とのつながり
同じブラウン大学のグループは2016年、慣れない場所で眠ると脳の左半球が「見張り番」のように警戒を続けることを報告していました(Tamaki et al., 2016)。
今回の研究は、その警戒モードの背景に、興奮系の物質が下がりきらない状態があるのかもしれない、という手がかりを与えてくれます(ただし、2つの研究のつながりを直接確かめたわけではありません)。くわしくは第一夜効果の記事もどうぞ。
04まとめ
旅先で寝つけないのは、気のせいではなく、脳がまだ「警戒モード」でアクセルをゆるめきれていないからかもしれません。次に枕が変わる夜は、「無理に寝よう」と力むより、脳の興奮を少しずつ下ろしていくイメージで過ごしてみてください。

Twenty healthy young adults each took two afternoon naps while special scans measured chemicals in the front part of the brain.
As expected, people took longer to fall asleep during the first nap. During the second, easier nap, the level of Glx, a group of chemicals that excite nerve cells, dropped as people moved from waking to sleep. In the first nap, it did not. Meanwhile, GABA, a chemical that calms nerve cells, did not clearly change in either nap.

Most people sleep badly on their first night in a strange place. This first-night effect has been documented since the 1960s, and earlier Brown University work showed that one half of the brain stays more watchful on such nights. In a study in the Journal of Neuroscience, a Brown University team has now used the effect as an experimental tool.
Twenty healthy young adults took two afternoon naps. Polysomnography tracked their sleep stages, while magnetic resonance spectroscopy measured two signals in the medial prefrontal cortex. One was Glx, an excitatory signal combining glutamate and glutamine. The other was GABA, the brain's main inhibitory chemical.
As expected, falling asleep took longer in the first session. In the second, Glx fell as they drifted into sleep, but in the first it did not. Across sessions, bigger drops in Glx went with falling asleep faster, and the Glx change statistically accounted for the gap between sessions.
GABA showed no clear change. The sample was small, naps differ from a night's sleep, and the scans show average levels across a region instead of activity at synapses. If the result holds, falling asleep may depend more on turning excitation down than on turning inhibition up.
