リチウムと聞くと、スマホの電池を思い浮かべる人が多いかもしれません。
でも実はリチウムは、私たちの体や脳の中にもごく微量に存在する金属です。そして2025年、アメリカ・ハーバード大学の研究チームが、この小さな金属がアルツハイマー病の始まりに関わっているかもしれない、という研究を発表しました。
0127種類の金属の中で、減っていたのはリチウムだけ
すると、調べた金属のうち、MCIの人の脳ではっきり減っていたのはリチウムだけでした。
さらにアルツハイマー病の脳では、脳にたまるアミロイドβ(ベータ)の塊(プラーク)がリチウムをくっつけて取りこんでしまい、脳の細胞が使えるリチウムがますます減っていることも分かりました。
02マウスでリチウムを減らすと、病気のような変化が加速
次に研究チームは、マウスで確かめました。エサからリチウムを減らし、脳のリチウムを約半分にすると、
- アミロイドβのたまりやタウタンパク質の異常が増えた
- 脳の免疫細胞(ミクログリア)が炎症モードになった
- 神経のつながりや配線のカバー(ミエリン)が失われた
- 記憶力の低下が早まった
という変化が起きました。アルツハイマー病のマウスだけでなく、ふつうに年をとったマウスでも似た影響が見られています。

03プラークに「捕まらない」リチウムで補うと…
研究チームはさらに、プラークにくっつきにくい「オロチン酸リチウム」という種類のリチウムを、とても少ない量でマウスに与えました。すると、病気の変化や記憶の低下が防がれ、すでに進んでいた変化が改善するケースも見られました。
実は、リチウムは昔から躁うつ病(双極性障害)の薬として使われてきました。ただ、その量は今回マウスで使われた量よりずっと多く、腎臓などへの副作用にも注意が必要です。また過去には、水道水のリチウム濃度が特に高い地域で認知症が少ない傾向があった、というデンマークの観察研究もあります。
ちなみに、私たちは飲み水や食べ物から、ふだんからごく微量のリチウムをとっています。体の中のリチウムが、どんな役割を果たしているのかは、まだよく分かっていない部分が多いのです。
04まとめ
アルツハイマー病の研究は、これまでアミロイドβやタウが主役でした。そこに「リチウム不足」という新しい視点が加わったのが、今回の研究の大きな意味です。
血液検査でリチウムの量を測れば、早い段階で見つけられるかもしれない――研究チームはそんな可能性も考えています。人での臨床試験の結果が出るまで、期待しつつ、焦らず待ちましょう。

When a Harvard Medical School team measured 27 metals in post-mortem brain tissue, one stood out. Lithium was the only metal significantly reduced at the stage of mild cognitive impairment (MCI). The study, published in Nature in 2025, compared brains from people with normal cognition, MCI, and Alzheimer's disease.
For decades, research on Alzheimer's has focused on two proteins, amyloid beta and tau. The new work suggests that a third factor, lithium deficiency, may also play a role. Amyloid plaques appeared to bind lithium, leaving less for brain cells to use.
To test cause and effect, the team cut brain lithium in mice by half through their diet. Plaque and tau changes sped up. Immune cells shifted into an inflammatory state, myelin and nerve connections were damaged, and memory declined faster. A very low dose of lithium orotate, a form that plaques capture less easily, prevented these changes.
The human evidence is thinner. It rests on a few hundred brains, and tissue examined after death cannot show whether low lithium is a cause or a consequence of the disease. No safe and effective dose for people has been established, and the researchers warn against taking lithium on your own.
